The Accuris One-Step RT-PCR Kit is a single-tube cDNA synthesis + PCR amplification kit for RNA → DNA → amplification workflows. The PR1100-50 is the 50-reaction pack — supports a meaningful validation or routine workflow batch. Per the Accuris Life Science Reagents Catalog: *"Convenient cDNA synthesis and subsequent PCR in a single tube."* The kit's chemistry is optimized for low-copy transcripts — the workflow is sensitive enough to detect dilute mRNA inputs without separate cDNA synthesis + PCR setup steps. Hot Start Taq Polymerase is included to prevent the non-specific amplification that two-step RT-PCR workflows typically suffer from at the PCR-setup transition.
| SKU | PR1100-50 |
| Type | Single-tube One-Step RT-PCR kit |
| Pack quantity | 50 reactions |
| Starting material | RNA |
| Workflow | cDNA synthesis (RT step) → PCR amplification (PCR step) in single tube — no transfer between steps |
| Sensitivity | Optimized chemistry for low-copy transcript detection |
| Kit contents | 1 tube of 20X thermostable RTase (RNase inhibitor included) + 1 tube of 2X One-Step Mix (PCR enhancers, 2 mM dNTPs, 6 mM MgCl₂) |
| Hot Start chemistry | Hot Start Taq Polymerase included — prevents nonspecific amplification at the RT-to-PCR transition |
| RNase inhibitor | Included in the 20X RTase tube — protects RNA from nuclease degradation during the RT step |
| Sister SKUs | PR1100-100 (100 reactions, $549), PR1100-S (sample, 10 reactions, $65) |
| List price | $304 |
Traditional two-step RT-PCR workflow:
One-step workflow (this kit) collapses all three into one tube: all reagents — RTase, Hot Start Taq, RNase inhibitor, buffer, dNTPs, primers, and RNA — are combined upfront, and the thermal cycler protocol drives both steps (an initial RT-temperature incubation for cDNA synthesis, then standard PCR cycling once the Hot Start activates the Taq). Because there is no tube transfer, the workflow eliminates the contamination, loss, and pipetting-error risk of the inter-step transfer.
Workflow advantages of one-step:
Hot Start Taq inclusion is critical for one-step success — without hot-start chemistry, the Taq would be active during the RT-temperature incubation and would generate non-specific PCR products from the as-synthesized cDNA before the formal PCR cycling begins.
| SKU | Reactions | List price | Per-reaction cost |
| PR1100-S | 10 | $65 | $6.50 |
| PR1100-50 (this product) | 50 | $304 | $6.08 |
| PR1100-100 | 100 | $549 | $5.49 |
Pick the PR1100-50 when: workflow throughput is moderate — typical clinical-research RT-PCR workflow consuming 50 reactions across a few weeks of project work, gene-expression studies running 50-sample panels, or method-development workflows beyond the 10-reaction sample scope. Step up to PR1100-100 for 100+ reactions per quarter; step down to PR1100-S for protocol validation.
Per the Accuris Life Science Reagents Catalog, One-Step RT-PCR Kit applications include multiplex RT-PCR (multiple primer pairs in one tube for multi-target gene-expression analysis) and gene-expression analysis (quantitative or semi-quantitative mRNA measurement at endpoint-RT-PCR scope; for fluorescence-based real-time RT-qPCR, see the qMAX RT-qPCR family).
Typical application contexts include pathogen detection (viral RNA detection, research-use only per kit positioning), gene-expression studies comparing target-gene mRNA levels across treatment and control conditions, mRNA-target validation confirming target presence in tissue or cell-line samples, and method-development RT-PCR for downstream qPCR assay design.
Routine gene expression analysis — the workhorse application. Endpoint RT-PCR for mRNA detection / semi-quantitative analysis.
Multiplex RT-PCR panels — multi-target gene expression measurement on a single sample input.
Pathogen RNA detection (research-use only) — viral RNA detection workflows in research labs.
Clinical-research mRNA workflows — clinical-research labs running RT-PCR for biomarker discovery or validation studies.
Method-development for downstream qPCR — protocol-development RT-PCR workflows that validate primer design before stepping to qMAX qPCR fluorescence chemistry.
Educational laboratory RT-PCR exercises — molecular-biology coursework introducing RT-PCR workflow.
Per-reaction workflow consumable. Each One-Step RT-PCR reaction consumes proportional reagent from the included tubes. The 50-reaction pack supports 50 RT-PCR reactions before reorder.
Storage. RT-PCR reagent storage applies — typically -20 °C; thermostable RTase storage protocol per product insert. RNase-free handling required throughout.
Pair with RNA template + sequence-specific primers. The kit includes the polymerase + RT + buffer + dNTPs + RNase inhibitor; operator supplies the RNA template and the gene-specific PCR primers (for one-step RT-PCR, the same primer pair drives both the RT step and the PCR step — no separate random hexamer / oligo-dT priming).
Confirm at quote: workflow throughput (drives 50-rxn vs 100-rxn pack-size selection), evaluation interest (PR1100-S sample at $65 for first-use validation), workflow application (gene expression vs pathogen detection vs method-development), and lead time.
Warranty: Per Accuris standard reagent terms — confirm at quote.
