Why Red Dye — Direct Gel Loading Workflow

The standard PCR-to-gel workflow has two pipetting steps after thermal cycling:

  1. Add loading dye to PCR product (typical 1–2 µL of 6X loading dye per 10–20 µL PCR product)
  2. Load mixture onto gel

The Red Dye master mix (PR1001-HSR family) eliminates step 1 — the loading dye is already in the PCR product because it was part of the 2X master mix from the start. After thermal cycling, the operator can load the PCR product directly onto the gel without a separate loading-dye addition step.

Workflow benefit:

  • One fewer pipetting step per reaction → reduces per-reaction handling time
  • Eliminates one source of pipetting error → improves workflow reproducibility
  • Reduces consumable load → no separate loading-dye reagent + tip consumption
  • High-throughput screening → meaningful when running 100+ PCR products per day

Trade-off: The Red Dye is inert during PCR — it does not affect the polymerase activity or PCR product integrity per Accuris design. However, some downstream workflows (sequencing, cloning, fluorescence-based detection) may not be compatible with PCR products containing the loading dye.
