SKU: PR1000-HF-1000

The PR1000-HF-1000 is the larger pack of the Accuris High Fidelity DNA Polymerase family — 1,000 units of HF polymerase at 2 U/µL for production-throughput cloning, sequencing template prep, mutagenesis, and microarray workflows. Right-sized for cloning core facilities, sequencing-prep production benches, and mutagenesis-production workflows where the smaller PR1000-HF-200 pack is too small for the rotation cycle. Each kit ships with 1 tube of HF Polymerase (500 µL at 2 U/µL = 1,000 units) + 5X buffer that — unique among the PR1000 family — already includes 5 mM dNTPs plus PCR enhancers and 15 mM MgCl₂.

Pack-Size Per-Unit Economics

SKUUnitsList pricePer-unit costPer-unit savings vs PR1000-HF-200
PR1000-HF-200200$175$0.875 / unit(baseline)
PR1000-HF-1000 (this product)1,000$652$0.652 / unit~25% lower

The 1,000-unit pack delivers ~25% per-unit savings vs the 200-unit pack — meaningful for production-scale HF workflows where the per-reaction enzyme cost is non-trivial.

A 1,000-unit pack supports approximately ~500 standard PCR reactions at 2 units per 50 µL HF reaction — typical 2–4 month rotation for a cloning-production bench running daily HF PCR.

When to Pick This Pack

Order PR1000-HF-1000 when:

  1. Cloning core facility production — academic core facilities or biotech cloning teams running HF PCR daily for ongoing cloning projects
  2. Sequencing-template-prep at volume — facilities running HF PCR for Sanger or NGS amplicon-based sequencing template prep at production scale
  3. Mutagenesis production workflow — site-directed mutagenesis production benches where every reaction must preserve all positions except the targeted site
  4. Microarray template prep — facilities producing microarray templates at production scale
  5. Antibody-engineering / protein-engineering workflows — workflows generating cloned variants of expression constructs at production scale where fidelity matters
  6. Step-up from PR1000-HF-200 — labs that started on the smaller pack and have established sufficient workflow throughput to justify the larger pack

Why HF Polymerase

Standard Taq DNA Polymerase has a measurable error rate during DNA synthesis. For workflows where individual PCR product molecules will be cloned, sequenced, or used as functional templates, the polymerase error rate matters because errors accumulate into the cloned, sequenced, or functional output. HF polymerases reduce error rate by 10–100× vs standard Taq through 3'→5' exonuclease proofreading activity that detects and corrects misincorporation errors during synthesis.

Blunt-End PCR Products

HF polymerase produces blunt-end PCR products — suitable for blunt-end cloning vectors. Standard Taq adds a 3'-terminal A overhang for TA-cloning compatibility; HF polymerase lacks this activity because the proofreading exonuclease removes terminal mismatches.

Workflow implication:

  • Blunt-end cloning vectors — direct ligation of HF PCR products
  • TA-cloning vectors — require post-PCR A-tailing step (separate Taq or A-tailing reagent)
  • Restriction-site cloning — restriction-digest the PCR product first; HF polymerase choice doesn't affect this

Buffer Formulation — dNTP Inclusion

The HF buffer already includes 5 mM dNTPs (1 mM in the 1X final reaction) — distinct from the PR1000 and PR1000-HS buffers, which require separate dNTPs. This simplifies reaction setup (no separate dNTP pipetting step), standardizes the dNTP final concentration at 1 mM (appropriate for high-fidelity PCR), and simplifies procurement (no separate dNTP line for HF workflows).

For workflows needing different dNTP concentrations or specialty dNTPs (modified bases, fluorescent-label dNTPs), supplemental dNTPs are needed.

Application Profile (per Brochure)

Per the Accuris Life Science Reagents Catalog, HF polymerase applications include blunt-end cloning (direct ligation of HF PCR products into blunt-end vectors), colony PCR where downstream use requires fidelity, fast PCR of short amplicons with reduced cycle times, GC- and AT-rich DNA amplification where proofreading helps with challenging-composition templates, and low-copy PCR of low-input templates. Mutagenesis and microarray applications are explicitly highlighted in the brochure as ideal uses.

Standalone Enzyme vs HF Master Mix

FormatWhen to pick
PR1000-HF-1000 (this product) — standalone enzymeCustom-buffer workflows, supplemental MgCl₂ workflows, cost-control on enzyme volume
PR1001-HF-200 / PR1001-HF-500 — HF 2X Master MixPre-formulated convenience for moderate-throughput
PR1001-HFHS-200 / PR1001-HFHS-500 — HF + Hot Start combinedChallenging templates needing both proofreading and hot-start specificity

For production-throughput cloning workflows, the standalone enzyme + buffer kit (this SKU) is often preferred over the master-mix format because it allows custom MgCl₂ adjustment for difficult templates.

Ordering & Configuration

dNTP independence. Unlike PR1000 / PR1000-HS, dNTPs are pre-formulated in HF buffer — no separate dNTP procurement.

Storage & handling. Per Accuris kit instructions — typical HF polymerase storage at −20 °C with limited freeze-thaw cycle tolerance.

Cloning-vector compatibility. Blunt-end products work directly with blunt-end vectors; TA-cloning workflows need post-PCR A-tailing.

Returns. Per the universe price-list category header: reagent products including PCR enzymes are non-returnable due to perishable nature.

Confirm at quote: downstream workflow (cloning / sequencing / mutagenesis / microarray fit), pack-size match (200 vs 1,000 units), standalone enzyme vs master-mix preference, cloning-vector format, shipping conditions (cold-chain), and lead time.

Warranty: Per Accuris Life Science Reagents standard kit warranty terms — confirm at quote.

Accuris PR1000-HF High Fidelity Polymerase family:

- PR1000-HF-200 — 200 units (smaller pack)

- PR1000-HF-1000 (this product) — 1,000 units (production-throughput pack)

- PR1001-HF-* — HF 2X Master Mix family (pre-formulated convenience)

- PR1001-HFHS-* — HF + Hot Start combined master mix family

Pick the 1,000-unit pack for production-throughput HF workflows where the per-unit savings matter and pack rotation supports the volume.
