Common Application — MTT Cell-Viability Assay

The MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay measures cell viability through metabolic reduction of the soluble yellow MTT to insoluble purple formazan crystals by viable cells (the reduction is performed by mitochondrial succinate dehydrogenase plus other reductases). Formazan crystals are dissolved in DMSO or isopropanol and read at 570 nm absorbance (with optional 630-690 nm reference subtraction).
