Per brochure: *"1X TBST contains 0.1% Tween-20."* The base TBS formulation is identical to EB1202 (137 mM NaCl, 2.7 mM KCl, 50 mM Tris) — the difference is the addition of 0.1 % Tween-20.

Why TBS-T for Western-Blot Membrane Wash

Western-blot workflows transfer SDS-PAGE-separated proteins onto nitrocellulose or PVDF membranes, then probe with primary and secondary antibodies to detect target proteins. Between each protocol stage (after blocking, after primary antibody, after secondary antibody), multiple wash steps remove unbound antibody from the membrane.

Tween-20 at 0.1 % in the wash buffer:

  • Disrupts hydrophobic interactions between antibodies and the membrane surface
  • Reduces non-specific antibody binding to membrane background (lower background signal)
  • Maintains specific antibody-antigen binding at the target band
  • Improves signal-to-noise ratio in the final detection (chemiluminescence, fluorescence, or chromogenic)

The 0.1 % concentration in TBS-T is 2× the 0.05 % concentration in PBS-T (EB1201) — Western-blot membrane wash typically requires more aggressive detergent action than ELISA plate wash because membranes have more hydrophobic surface area than polystyrene plates and benefit from stronger detergent disruption.
