qPCR Plate Sealing: Why Optical Clarity Matters

qPCR thermal cycling has two stages where seal optical properties matter:

Without optical-clear film, standard foil seals block fluorescence, qPCR cannot read through them, and removing seals before reading wastes qPCR throughput, introduces evaporation losses, and risks cross-well contamination.

Recommended Sealing Setting

Per SmartSeal brochure: 170 °C, 2.5 seconds for "Most Clear-Seals" (the recommended starting setting for clear sealing films like MS1000-PCR1). Actual settings may need adjustment per:

• PCR plate brand and material. Different PCR plate manufacturers may have slightly different surface chemistry
• PCR plate skirting. Fully skirted vs semi-skirted vs non-skirted plates may seal slightly differently
• Plate temperature at seal time. Pre-cooled plates may require slightly longer seal time
