The F1000-SD-100 is the 100-assay Accuris ssDNA Quantification Kit — the single-stranded DNA-specific chemistry in the F1000 fluorometric family. Distinct from the dsDNA-binding chemistry of the F1000-BR / F1000-HS variants (which don't reliably detect single-stranded DNA), the F1000-SD uses a chemistry that binds ssDNA specifically — useful for sequencing template prep (cycle-sequencing extension products are single-stranded), oligonucleotide quantification, denatured-DNA workflows, and any application where the target nucleic acid is single-stranded.

Why ssDNA-Specific Chemistry

dsDNA-binding chemistry (Qubit BR / HS, F1000-BR / F1000-HS) binds the dsDNA major groove and produces fluorescence proportional to dsDNA concentration. Single-stranded DNA doesn't have the dsDNA major-groove geometry — dsDNA-binding chemistry produces little or no signal on ssDNA samples, undercounting or missing the ssDNA entirely.

The F1000-SD uses an ssDNA-binding dye that detects single-stranded DNA specifically. This matters for:

  • Cycle-sequencing extension product quantification — Sanger cycle-sequencing produces ssDNA extension products that need quantification before sequencer loading
  • Oligonucleotide quantification — synthetic oligonucleotides (PCR primers, hybridization probes, antisense oligos) ship as ssDNA and require ssDNA-specific quantification
  • Denatured DNA workflows — DNA samples that have been heat-denatured or chemical-denatured to single-stranded form
  • Bisulfite-converted DNA — methylation-analysis workflows where bisulfite conversion produces single-stranded DNA
