The chassis-controlled block setpoint temperature is not always identical to the actual sample temperature. Sample temperature lags the block during the heat-up ramp, larger sample volumes take longer to equilibrate, different tube wall thicknesses produce different thermal lag, well-fit versus loose-fit block-to-tube contact affects heat transfer, and equilibration depends on the operating temperature relative to ambient.
